 岐阜県御嶽山の土壌から採取した菌核において,菌核形成菌を推定するためにクローンライブラリー法および菌類の分離をおこなった。クローンライブラリー法により,菌核形成菌として報告されているCenococcum geophilum が認められた。分離された糸状菌は,その大部分が子囊菌類であった。培養法ではC. geophilumが認められなかったものの, DSE に類別されると考えられる未同定菌3 株が,人工培地上で菌核を形成した。本研究で供試した菌核は, C. geophilum だけでなく,他の菌種により形成された可能性もあると考えられた。

Streptomyces parvulus 2297, which is a host for site-specific recombination according to actinophage R4, is derived from the type strain ATCC 12434. Species of S. parvulus are known as producers of polypeptide antibiotic actinomycins and have been considered for industrial applications. We herein report for the first time the complete genome sequence of S. parvulus 2297.

We investigated butanol recovery by pervaporation separation, using a silicalite-1 membrane, from batch cultures of butanol-producing Clostridium beijerinckii SBP2 grown on sweet sorghum juice as a fermentation medium. The pervaporation system yielded 73% (w/v) butanol from intact feed cultures containing 1% (w/v) butanol, and had a butanol
permeation flux of 11 g mL2 hL1. Upon neutralization and activated charcoal treatment of the feed cultures, butanol
yield and total flux increased to 82% (w/v) and 40 g mL2 hL1
, respectively. This system is applicable to refining processes for practical biobutanol production from a promising energy crop, sweet sorghum.

A dairy wastewater treatment system composed of the 1st segment (no aeration) equipped with a facility for the destruction of milk fat particles, four successive aerobic treatment segments with activated sludge and a final sludge settlement segment was developed. The activated sludge is circulated through the six segments by settling sediments (activated sludge) in the 6th segment and sending the sediments beck to the 1st and 2nd segments. Microbiota was examined using samples from the non-aerated 1st and aerated 2nd segments obtained from two farms using the same system in summer or winter. Principal component analysis showed that the change in microbiota from the 1st to 2nd segments concomitant with effective wastewater treatment is affected by the concentrations of activated sludge and organic matter (biological oxygen demand [BOD]), and dissolved oxygen (DO) content. Microbiota from five segments (1st and four successive aerobic segments) in one location was also examined. Although the activated sludge is circulating throughout all the segments, microbiota fluctuation was observed.

An endohyphal bacterium (strain B1-EBT) living in association with the fungus Mortierella elongata FMR23-6 I-B1 was isolated from the fungal cell homogenate and studied for its taxonomic allocation. Cells were Gram-stain-negative, rod-shaped, non-spore-forming, non-motile, and negative for oxidase and catalase. Strain B1-EBT required cysteine for growth and grew at temperatures between 4-35 ºC. A comparative analysis of 16S rRNA gene sequences revealed that strain B1-EBT forms a distinct clade in the family Burkholderiaceae, encompassing a group of endosymbionts associated with several soil isolates of M. elongate.

Newly exposed volcanic substrates contain negligible amounts of organic materials. Heterotrophic organisms in newly formed ecosystems require bioavailable carbon and nitrogen provided by pioneer microbes (e.g., photo- and chemolithotrophs), which fix atmospheric CO2 and N2. However, knowledge of the developmental mechanisms of initial ecosystems, especially the association between microbial succession and environmental change, is still limited. This study reports the unique process of microbial succession in fresh basaltic ash, which is affected by long-term exposure to volcanic sulphur dioxide gas (SO2).

The response of the soil food web structure to soil quality changes during long-term anthropogenic disturbance due to farming practices has not been well studied. We evaluated the effects of three tillage systems: moldboard plow/rotary harrow (MP), rotary cultivator (RC), and no-tillage (NT), three winter cover-crop types (fallow, FL; rye, RY; and hairy vetch, HV), and two nitrogen fertilization rates (0 and 100 kg N ha−1 for upland rice, and 0 and 20 kg N ha−1 for soybean production) on changes in nematode community structure.

A halo- and organo-sensitive oligotrophic bacterium, designated strain SY-6T, was isolated from humus forest soil at Gyeryong mountain in Korea. The strain was Gram-negative, strictly aerobic, non-motile and formed yellow-pigmented rod colonies on 100-fold diluted nutrient broth. Strain SY-6T grew between pH 6.0 and 7.0 (optimal growth at pH 7.0), between 10 and 37 °C (optimal growth at 28 °C) and at salinities of 0–0.5 % (w/v) NaCl, growing optimally at 0.01 % (w/v) NaCl. On the basis of 16S rRNA gene sequence analysis, strain SY-6T was shown to belong to the genus Sphingomonas and showed closest phylogenetic similarity to Sphingomonas polyaromaticivorans B2-7T (96.7%). The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, phosphatidylcholine and sphingoglycolipid. The predominant ubiquinone and polyamine components were Q-10 and sym-homospermidine, respectively. The major fatty acids were C18:1 ω7c (56.9%) and C16:0 (10.1%). The DNA G+C content of this novel isolate was 65.3 mol%. On the basis of polyphasic evidence from this study, strain SY-6T represents a novel species of the genus Sphingomonas for which the name Sphingomonas oligoaromativorans sp. nov. is proposed. The type strain is SY-6T (=KACC 12948T=NBRC 105508T).

The 2000 eruption of Mount Oyama in the Island of Miyake (Miyake-jima) created a unique opportunity to study the early ecosystem development on newly exposed terrestrial substrates. In this study, bacterial and fungal communities on 9- and 11-year-old volcanic deposits at poorly to fully vegetation-recovered sites in Miyake-jima, Japan, were characterized by conventional culture-based methods and pyrosequencing of 16S rRNA and 18S rRNA genes.

A thallium-tolerant, aerobic bacterium, designated SK200a-9T, isolated from a garden soil sample was characterized by polyphasic approaches. From the results, the new genus and species, Alsobacter metallidurans gen. nov., sp. nov. is proposed for strain SK200a-9T in the order Rhizobiales. The type strain is SK200a-9T (=NBRC 107718T =CGMCC 1.12214T).

The fungus Mortierella elongata FMR23-6 harbors an endobacterium inside its mycelium. Attempts to isolate the endobacterium from the fungus were not yet successful, but a highly purified bacterial fraction was prepared. Here, we report the draft genome sequence of the endobacterium.

1910~40年代、アメリカの土壌細菌学者H.J. Connは、直接検鏡法や計数培地を開発し、土壌細菌のslow growerに着目した研究を展開して、土壌微生物学と細菌分類学の双方に貢献した(例えば、Conn, 1914; Conn, 1917)。S.N. Winogradskyが考えたautochthonous floraの概念も、元をたどればConnの研究に行き着く。Connが追究したslow growerの一部はArthrobacter globiformisとして記載され(Conn and Dimmick, 1947)、土壌に固有な細菌の重要な一つとして位置づけられてきた。Connは、純粋培養を基盤にして、分類学や生理・生化学などの観点から土壌微生物研究を展開させた点で重要な功績を残したと言える。現在、微生物学は純粋培養の菌株(strain)からゲノムに基盤がシフトしている。このシフトによって、メタゲノミクスが始まり、土壌細菌群集の知識は大きく広がった。本稿では、このような研究動向に触れながら、筆者らが行ってきた土壌細菌の分類および群集解析に関する研究を紹介する。

However, the effects of their interaction are less well known, particularly in Asian countries. 土壌中で起こる硝化と脱窒は細菌のエネルギー代謝であり、グローバルな窒素循環の駆動につながっている。土壌の構造と水分含量は、硝化と脱窒の活性に影響を及ぼし、N2Oが副産物として発生する。糸状菌においてもN2Oを産物とする脱窒活性がわかってきた。実際の土壌でのN2O生成の主役が細菌なのか糸状菌なのか?その探究が始まっている。

An anammox assay involving a 15N tracer and gas chromatography-mass spectrometry revealed that the potential
anammox activity accounted for 1 to 5% of total N2 production in a ravine paddy field, Japan. Among four 4-cm-deep
layers, the top layer showed the highest activity. Clone libraries showed that the DNA in the top layer contained
sequences related to those of Candidatus ‘Brocadia fulgida’, Ca. ‘B. anammoxidans’, and Ca. ‘Kuenenia stuttgartiensis’.
These results suggest that a specific population of anammox bacteria was present in paddy soils, although a small part
of dinitrogen gas was emitted from the soil via anammox.

Microbial colonization on new terrestrial substrates represents the initiation of new soil ecosystem formation. In this study, we analyzed the early bacterial communities growing on volcanic ash deposits derived from the 2000 Mount Oyama eruption in the Island of Miyake (Miyake-jima), Japan. A site was established in an unvegetated area near the summit and investigated over a 6-year period from 2003 to 2009. Collected samples were acidic (pH 3.0-3.6), did not utilize any organic substrates in ECO microplate assays (Biolog, Inc.), and harbored around 106 (g dry weight) −1 of autotrophic Fe(II) oxidizers by most-probable-number counts. Acidithiobacillus ferrooxidans, Acidithiobacillus ferrivorans, and the Leptospirillum groups I, II and III were found to be abundant in the deposits by clone library analysis of bacterial 16S rRNA genes. The numerical dominance of At. ferrooxidans was also supported by analysis of the gene coding for the large subunit of the form I ribulose 1,5-bisphosphate carboxylase/oxygenase (RubisCO). Comparing the 16S rRNA gene clone libraries from samples differing in age, shifts in Fe(II)-oxidizing populations seemed to occur with deposit aging. The detection of known 16S rRNA gene sequences from Fe(III)-reducing acidophiles promoted us to propose the acidity-driven iron cycle for the early microbial ecosystem on the deposit.

初成土壌環境における微生物-三宅島土壌生態系再生のメカニズムを探る-

森林をはじめとする植物-土壌生態系では、光合成による一次生産と微生物による有機物分解のバランスが成り立ち、豊かな生物相が維持されている。しかし、火山噴火というイベントはこの生態系を壊してリセットしてしまう。新たに生じた火山灰などの火山砕屑物や溶岩に住み始める生物は、肉眼では見えない微生物である。本稿では、三宅島2000年噴火火山灰堆積物に住みつく微生物について、2003年から6年間にわたって調査してきた結果を紹介する。まず、調査初年時に採取した火山灰堆積物の細菌密度の測定結果では、すでに1gあたり108の高いレベルに達していた。直接試料から抽出したDNAについて16SリボソーマルRNA遺伝子を解析した結果から推定した細菌群集の解析からは、レプトスピリリウム・フェロオキシダンス(Leptospirillum ferrooxidans)やアシディチオバチルス・フェロオキシダンス(Acidithiobacillus ferrooxidans)といった独立栄養性の鉄酸化細菌が優占することがわかった。供試火山灰堆積物にはCO2吸収活性があり、十分に高いニトロゲナーゼ活性も検出されており、これらの活性は鉄酸化細菌に由来することが推察された。2009年の調査においても、三宅島雄山上部の火山灰堆積物は酸性状態に維持され、鉄酸化細菌の優占が続き、化学合成無機独立栄養が中心の微生物生態系であることが示唆された。以上の結果をもとに、火山灰堆積物に形成される微生物生態系のエネルギー代謝と初成土壌への有機・無機物質の蓄積について推察する。

Ethanol was efficiently produced from three varieties of sweet sorghum using repeated-batch fermentation without pasteurization or acidification. Saccharomyces cerevisiae cells could be recycled in 16 cycles of the fermentation process with good ethanol yields. This technique would make it possible to use a broader range of sweet sorghum varieties for ethanol production.

糸状菌細胞に内生する細菌の存在とその検出法

近年,糸状菌の細胞内部に内生する細菌の検出例が報告されている。例えば,Rhizopus属菌の菌糸内部に分布する内生細菌は,これまでRhizopus属菌が生成すると考えられていたリゾキシンを生成することが明らかとなった。このような背景から今後、糸状菌を扱う研究全般において内生細菌の検出が重要な項目のひとつになると考えられる。本稿では,糸状菌細胞内生細菌の検出方法として,グラム陰性細菌の細胞壁成分のひとつであるエンドトキシンの定量による内生細菌の検出法,細菌16S rRNA遺伝子を標的としたPCR法による内生細菌の検出,蛍光顕微鏡および透過電子顕微鏡を用いて,菌糸内に分布する内生細菌を直接観察する方法について,筆者らの手法を中心に既往研究を加えて解説した。本稿で紹介した検出方法は透過電子顕微鏡観察を除いて,比較的簡易であることから,糸状菌の細胞内生細菌の分布調査に広く活用されることが期待される。糸状菌細胞内生細菌および共生体(共存体)の土壌における生態学的役割については今後の研究課題である。

A Gram-negative, aerobic, heterotrophic bacterium able to use thiosulfate (optimum concentration, 10 mM) as an additional energy source, designated KP1-19T, was isolated from a 22-year-old volcanic deposit at a site lacking vegetation on the island of Miyake, Japan. In this study, its taxonomic position was determined by a polyphasic approach. 16S rRNA gene sequence analysis indicated that strain KP1-19T is closely related to Limnobacter thiooxidans LMG 19593T, the single species in the genus Limnobacter within the class Betaproteobacteria. The level of 16S rRNA gene sequence identity between strains KP1-19T and LMG 19593T was 97.7%. The major cellular fatty acids in KP1-19T were found to be C16:1ω7c, C18:1ω7c, C16:0 and C10:0 3-OH, which are characteristic of the genus Limnobacter. The DNA-DNA relatedness of strains KP1-19T with LMG 19593T is 18%. Therefore, strain KP1-19T represents a novel species, for which the name Limnobacter litoralis sp. nov. is proposed; the type strain is strain KP1-19T (=LMG 24869T =NBRC 105857 T =CIP 109929T).

Ribosomal RNA (rRNA) sequence-based molecular techniques emerged in the late 1980s, which completely changed our general view of microbial life. Coincidentally, the Japanese Society of Microbial Ecology (JSME) was founded, and its official journal “Microbes and Environments (M&E)” was launched, in 1985. Thus, the past 25 years have been an exciting and fruitful period for M&E readers and microbiologists as demonstrated by the numerous excellent papers published in M&E. In this minireview, recent progress made in microbial ecology and related fields is summarized, with a special emphasis on 8 landmark areas; the cultivation of uncultured microbes, in situ methods for the assessment of microorganisms and their activities, biofilms, plant microbiology, chemolithotrophic bacteria in early volcanic environments, symbionts of animals and their ecology, wastewater treatment microbiology, and the biodegradation of hazardous organic compounds.

Microscopic and molecular analyses showed the presence of endobacteria inside the mycelia of four out of twelve nitrous oxide (N2O)-producing fungal isolates identified as Mortierella elongata. The 16S rRNA genes were successfully amplified with directly extracted DNA from the endobacterium-containing fungal strains and all of their sequences were related to that of Candidatus Glomeribacter gigasporarum in the family Burkholderiaceae. The bacterial endotoxin was detected in the endobacterium-positive fungal strains but the trace levels in endobacterium-negative strains. No significant relationship was found between the fungal N2O-producing activity and the presence of endobacteria.

16S rRNA遺伝子解析が導入される前の時代,土壌細菌研究者の多くは,分離菌株の分類学的位置づけがどのような生態的意味をもつかについて一度は考え悩む時があったと思う.そのような時代の最初の頃に,栗原は「ecological species(生態学的種)の概念の確立」を説いた(1965年).2000年代に入り,遺伝子変異の観点から細菌のecotype(エコタイプ)や細菌種の見直しが盛んに議論されている.ここでは,まず栗原の考察を読み解き,Cohanが提唱する細菌の「エコタイプ」を解説するとともに,著者らが扱ってきた初成土壌(三宅島2000年噴火火山灰堆積物)に住みつく細菌の分析結果を紹介して,細菌の生態学的種について考える話題を提供する.

This study aimed to characterize fungal communities in upland rice fields managed with tillage/non-tillage and winter cover-cropping (hairy vetch and cereal rye) practices, using PCR-based molecular methods. The study plots were maintained as upland fields for 5 years and the soils sampled in the second and fifth years were analyzed using T-RFLP (terminal restriction fragment length polymorphism) profiling and clone libraries with the internal transcribed spacer (ITS) region and domain 1 (D1) of the fungal large-subunit (fLSU) rRNA (D1fLSU) as the target DNA sequence. From the 2nd-year-sample, 372 cloned sequences of fungal ITS-D1fLSU were obtained and clustered into 80 nonredundant fungal OTUs (operational taxonomic units) in 4 fungal phyla. The T-RFLP profiling was performed with the 2nd- and 5th-year-samples and the major T-RFs (terminal restriction fragments) were identified using a theoretical fragment analysis of the ITS-D1fLSU clones. These molecular analyses showed that the fungal community was influenced more strongly by the cover-cropping than tillage practices. Moreover, the non-tilled, cover-cropped soil was characterized by a predominance of Cryptococcus sp. in the phylum Basidiomycota. We provided a genetic database of the fungal ITS-D1fLSUs in the differently managed soils of upland rice field.

土壌環境中に存在する微生物の多様性や土壌環境変化への微生物群集の応答を調べるため、リアルタイム定量PCRアッセイと末端制限断片長多型解析(T-RFLP)法を併用した手法を確立した。この解析手法から、標的遺伝子領域に基づく微生物群の末端制限断片(T-RFs)サイズとピーク高(相対値)の情報が得られる。T-RFLPプロファイリングの実測情報とクローン解析から得られた遺伝子配列のin silico解析によるT-RFsプロファイル情報がほぼ一致することから、本法が精度の高い解析技術であることを示した。また、検出されたT-FRsプロファイル情報をもとに多次元尺度構成法分析で微生物群集の類似性とその動態を表し、T-RFsの相対的存在量の頻度分布順位図から微生物の多様性を示す方法を紹介した。さらに、T-RFsプロファイル情報をもとに土壌微生物群集を推定するバイオインフォマテッィクスの利用方法を示した。今後のデータベースの充実により、定量PCR-T-RFLP法が土壌微生物群集モニタリングに大いに役立つことに期待している。

Twenty seven soil bacterial isolates able to grow on cinnamic acid or ferulic acid as the sole carbon and energy source were described for their ability to utilize other phenylpropanoids and their phylogenetic interrelationship. The 16S rRNA gene sequence analysis revealed that 26 of 27 soil isolates were closely related to Pseudomonas putida biovar A and classified into 6 OTUs: cinnamic acid utilization-positive (OTUs, CN-1 to CN-3) and -negative groups (FR-1 to FR-3). P. putida NBRC 14164T and JCM 13061 were belonged to FR-3. A probable reclassification of P. putida was discussed in relation to the utilization of phenylpropanoids.

We have assayed rates of atmospheric CO and hydrogen uptake, maximum potential CO uptake and the major phylogenetic composition of CO-oxidizing bacterial communities for a variety of volcanic deposits on Miyake-jima, Japan. These deposits represented different ages and stages of plant succession, ranging from unvegetated scoria deposited in 1983 to forest soils on deposits > 800 yr old. Atmospheric CO and hydrogen uptake rates varied from −2.0 ± 1.8 − 6.3 ± 0.1 mg CO m−2 d−1 and 0.0 ± 0.4 − 2.0 ± 0.2, mg H2 m−2 d−1, respectively, and were similar to or greater than values reported for sites on Kilauea volcano, Hawai'i, USA. At one of the forested sites, CO was emitted to the atmosphere, while two vegetated sites did not consume atmospheric hydrogen, an unusual observation. Although maximum potential CO uptake rates were also comparable to values for Kilauea, the relationship between these rates and organic carbon contents of scoria or soil indicated that CO oxidizers were relatively more abundant in Miyake-jima deposits. Phylogenetic analyses based on the large sub-unit gene for carbon monoxide dehydrogenase (coxL) indicated that many novel lineages were present on Miyake-jima, that CO-oxidizing Proteobacteria were prevalent in vegetated sites and that community structure appeared to vary more than composition among sites.

The PCR amplification-based analysis of microbial diversity is subject to potential problems. In this study, to minimize the bias toward a 1:1 ratio in multitemplate PCR, a real-time PCR assay was carried out using a quenching fluorescence dye primer and amplification efficiency was monitored. Then terminal-restriction fragment length polymorphism (T-RFLP) profiling was performed using the PCR product with minimized PCR bias. This method was applied to an analysis of the diversity of the archaeal community in an upland rice field under different tillage systems and winter cover cropping. Terminal restriction fragments (T-RFs) of PCR-amplified archaeal 16S rRNA genes were assigned to the gene sequences recovered from the same soil by using an archaeal 16S rRNA gene clone library. Our results indicated that soil archaeal members were not influenced but the relative abundance of archaeal species particularly those belonging to Crenarchaeota which changed between the tillage and non-tillage treatments.

The relationships between soil microbial properties and nitrous oxide emission were examined in upland soil under different tillage systems [no tillage (NT), rotary and plow tillage] and cover crop systems (fallow, cereal rye, and hairy vetch) in 2004 and 2005. Microbiological analyses included the determination of soil ergosterol as an indicator of fungal biomass, bacterial plate counting, and MPN estimations of ammonia oxidizers and denitrifiers. The combined practice of NT with rye-cover crop treatment increased fungal biomass but not bacterial populations in 0−10 cm deep soils. Such increase in fungal biomass was not found in 10−20 cm and 20−30 cm deep cover-cropped NT soil. The combined practice of NT with rye-cover cropping resulted in higher in situ N2O emission rates compared with rotary- and plow-till treatments. N2O flux was positively correlated with soil ergosterol content but not with denitrifier MPN and other soil chemical properties. These results suggested a significant contribution of fungi to N2O emission in cover-cropped NT soils.

The role of microbes in the early development of ecosystems on new volcanic materials seems to be crucial to primary plant succession but is not well characterized. Here we analyzed the bacterial community colonizing 22-year-old volcanic deposits of the Miyake-jima Island (Japan) using culture-based and 16S rRNA gene clone library methods. The majority of 91 bacterial isolates were placed phylogenetically in two clusters (A and B) of the Betaproteobacteria. Cluster A (82% of isolates) was related to the genus Limnobacter and Cluster B (9%) was affiliated with the Herbaspirillum clade. The clone library analysis supported the predominance of Cluster B rather than Cluster A. Strain KP1-50 of Cluster B was able to grow on a mineral medium under an atmosphere of H2, O2, and CO2 (85:5:10), and characterized by its large-subunit gene of ribulose 1,5-bisphosphate carboxylase/oxygenase (rbcL) and nitrogenase reductase gene (nifH). In contrast, strains of Cluster A did not grow chemolithoautotrophically with H2, O2, and CO2 but increased their cell biomass with the addition of thiosulfate to the succinate medium, suggesting the use of thiosulfate as an energy source. From phenotypic characterization, it was suggested that the Cluster A and B strains were novel species in the genus Limnobacter and Herbaspirillum, respectively.

Agricultural systems provide both sources and sinks of greenhouse gas (GHG), which includes carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O). This paper addresses soil organic carbon (SOC) storage and GHG uptake and emission in agriculture, emphasizing SOC changes associated with crop and tillage management and the contribution of microbial activities and farming practices to GHG fluxes. Cropland soils store great amounts of organic carbon and are one of the sinks for atmospheric CO2. Plant biomass and soils store about 500 and 1100 Pg C, respectively, on the global scale; carbon stored in soils is mainly in the form of soil organic matter. Conservation tillage, cover crop management, and manure application are recommended to enhance SOC storage. However, it is possible that management practices resulting in SOC sequestration could do so at the expense of increased amounts of other GHG emissions. Therefore, the trade-off relationship among SOC storage, GHG emission and recommended management practices should be monitored through long-term studies. GHG emission from cropland is strongly influenced by soil microbial activities, such as decomposition, CH4 production, nitrification, and denitrification; an alternative farming strategy is desirable to manage soil microbial diversity and activity for the mitigation of GHG emissions. In addition, CO2 emissions from farm machinery used to produce food for the growing world population would be another agricultural GHG source.

A soil column was percolated with Triton X-100 (octylphenol polyethoxylates; OPEOn) under aerobic conditions for 300 days and OPEOn-degrading bacteria were isolated and characterized phylogenetically. Populations of total culturable bacteria and OPEOn (1000 mg L−1)-resistant bacteria increased 10 times and 46 times, respectively, during the percolation performance. 16S rRNA gene sequence analysis of 128 OPEOn-resistant isolates revealed an increase in the population level of Alphaproteobacteria and decreases in those of Betaproteobacteria and Actinobacteria with the exposure to the compound. Four OPEOn-degrading strains were isolated and characterized as being related to Bradyrhizobium liaoningense (3 strains) and Afipia sp. (1 strain).

Thallium (Tl) is a heavy metal found in trace amounts in the earth’s crust and has been studied to a much lesser degree than other heavy metals. Since the discovery of high-temperature superconducting components in the Tl-Ca-Ba-Cu-O system, Tl has attracted greater attention as a potential major pollution source of the future. In this study, the response of soil cultivable communities to the addition of Tl was examined with a river sediment sample from a heavy metal-contaminated site and samples of uncontaminated garden and arable soils. Soil suspension experiments were performed to homogenize the soils to evenly distribute both the microbial populations and Tl and reduce spatial variability. From soil suspensions loaded with Tl at 0.24 to 0.98 mM, bacteria tolerant to 0.49 mM Tl were isolated and characterized phylogenetically. The background level of culturable Tl-tolerant bacterial populations was 0.0003-0.013% for the uncontaminated soils and 0.02 to 0.04 % for the contaminated river sediment sample. The Tl-tolerant fraction increased rapidly in response to Tl loading and dominated the bacterial community. Thallium-tolerant culturable bacteria isolated from the garden soil were classified into 9 groups of Alphaproteobacteria, those from the arable soil were identified as Pseudomonas spp., and those from the contaminated river sediment were Bacillus niacini.
